I have to extend that to "books" rather than just one book.
My choice is The Malazan Books of the Fallen by Steven Erikson - 12 in all so far in a continuous story and each volume a block-buster.

it is always the book I am reading now, but the one that had the biggest influance on my life was The Rune Staff, by Micheal Moorcock. It was the first book I had read in full in my life I was 17 at the time.

Watchmen. If you don't count graphic novels then Sirens of Titan.

Hmm...favorite books would be

- Love and Louis XIV: The Women in the Life of the Sun King by Antonia Fraser

It made me fall in love with the Sun King even more than before.

- Deep Love Ayu

This is a manga so I'm not sure if it counts. However if it does, I will never read anything as disturbing yet beautiful as this book! It changed my life.

- Any book by Erma Bombeck

She always makes my bad days better. I wish that she was still alive. She was far too young to die. Rest in peace, Erma. We all miss you and your wonderful sense of humor!

Kraken - China Mieville
Perdido Street Station - China Mieville
Footfall - Larry Niven and Jerry Pournelle
The Saga of Seven Suns - Kevin Anderson
Dune - Frank Herbert

I think Perdido Street Station really changed me as a person though.

So many to choose from but the series of books that I continue to go back to, for comfort and familiarity is the Belgariad and Mallorean series by David Eddings.
